Background:
  GM-CSF is produced in response to a number of inflammatory mediators by mesenchymal cells present in the hemopoietic environment and at peripheral sites of inflammation. GM-CSF is able to stimulate the production of neutrophilic granulocytes, macrophages, and mixed granulocyte-macrophage colonies from bone marrow cells and can stimulate the formation of eosinophil colonies from fetal liver progenitor cells. GM-CSF can also stimulate some functional activities in mature granulocytes and macrophages. GM-CSF receptors shows significant homologies with other receptors for hematopoietic growth factors , including IL2-beta, IL-3 , IL-6 , IL-7 , EPO and the Prolactin receptors

 

Description :
  Recombinant murine GM-CSF produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 125 amino acids and having a molecular mass of 14285.35 Dalton.
Murine GM-CSF is purified by proprietary chromatographic techniques.

 

Physical Appearance:
  Sterile Filtered White lyophilized (freeze-dried) powder.

 

Formulation:
  Murine GM-CSF was lyophilized after extensive dialysis against 2mM sodium phosphate buffer pH= 7.4╠0.1.
Solubility:
  It is recommended to reconstitute the lyophilized GM-CSF in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

 

Stability:
 
  Lyophilized GM-CSF although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution GM-CSF should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please avoid freeze-thaw cycles.

 

Purity:
  Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

 

Amino acid sequence:
  The sequence of the first five N-terminal amino acids was determined and was found to be Met-Ala-Pro-Thr-Arg.

 

Dimers and aggregates:
  Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

 

Biological Activity:
  ProSpec's Murine GM-CSF is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of the proliferation of murine FDC-P1 cell line is less then 0.2 ng/ml, corresponding to a Specific Activity of 5×10 6IU/mg.

 

Endotoxin:
  Less than 0.1 ng/µg (IEU/µg) of Murine GM-CSF .

 

Protein content:
  Protein quantitation was carried out by two independent methods:

1. UV spectroscopy at 280 nm using the absorbency value of 0.765 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (InliGenetics).
2. Analysis by RP-HPLC, using a standard solution of GM-CSF as a Reference Standard.

Background:
  The overall structure of GM-CSF is highly compact and globular with a predominantly hydrophobic core, is characterised by a 4-alpha-helix bundle. The helices are arranged in a left-handed anti-parallel fashion, with two overhand connections. Within the connections is a two-stranded anti-parallel beta-sheet. The tertiary structure has a topology similar to that of (pig) growth factor and interferon-beta. Most of the proposed critical regions for receptor binding are located on a continuous surface at one end of the molecule that includes the C terminus .

Quinidine是一种临床抗心律失常药,它影响心肌中的离子电流,并且已被证明是多种细胞类型中K+通道的几类强效阻断剂。浴奎尼丁的应用导致IK的峰值幅度的剂量依赖性降低。在0 mV的阻断IK的KD估计为41μM. Quinidine引起IK衰变的剂量依赖性增加,并且这种效应通过膜去极化得到增强。Quinidine还造成了5 mV的超极化的稳态失活曲线移位和增加复活时。Quinidine不影响灭活测量-发病30 MV 。

体内研究

奎尼丁被迅速吸收,口服后血浆浓度峰值60-90分钟。其他盐(葡萄糖酸盐,聚半乳糖酸盐)吸收较慢,峰值浓度较低。奎尼定与血浆蛋白的结合率约为70- 90%。它经历肝氧化代谢形成n -氧化物,3-羟基,o – dem乙基和2′-喹茚酮。超过一半的患者在治疗的第一年就停止服用奎尼丁因为副作用。这些症状通常包括腹泻、恶心和呕吐,这些症状不一定与血浆浓度高有关。奎尼丁抑制大鼠安非他明的代谢。奎尼丁预处理可使对-羟基安非他明在24小时、48小时、7.2小时和24.1%车辆控制水平下的排泄量显著降低,同时使安非他明在24小时至48小时、542%的控制水平下的排泄量显著增加。

经典实验操作(仅供参考)

动物实验
  • 奎尼丁溶解在50%乙醇中
  • 大鼠:3只大鼠被放在单独的代谢笼子里,可以自由获得食物和水。24小时后收集尿液(0小时),大鼠接受以下治疗:(1)不治疗;(2) 80 mg奎尼丁/kg (po) 1.0 mL/kg, 50%乙醇(1.0 mL/kg)。两小时后,所有3只大鼠均以1.0 mL/kg的剂量接受15毫克安非他命(po)。然后在服用奎尼丁后的24小时和48小时收集尿液。这个过程重复三次。收集尿液后,测量尿液体积和pH值,并储存至分析。
