Background:
  Oncostatin M （OSM） is a member of the interleukin-6 family of cytokines. Of these cytokines, OSM is closely related structually, genetically and functionally to leukemia inhibitory factor. However, OSM-specific biological activities have been reported in hematopoiesis and liver development.

 
Description :
  Recombinant Human OSM produced in E.Coli is a single, non-glycosylated polypeptide chain containing 227 amino acids and having a molecular mass of 26kDa.
Recombinant OSM is purified by proprietary chromatographic techniques.

 
Physical Appearance:
  Sterile Filtered White lyophilized （freeze-dried） powder.

 
Formulation:
  Lyophilized from a concentrated （1mg/ml） solution containg no additives.

 
Solubility:
  It is recommended to reconstitute the lyophilized rHuOSM in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

 
Stability:
 
  Lyophilized Recombinant Human OSM although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution OSM should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein （0.1% HSA or BSA）.
Please avoid freeze-thaw cycles.

 
Purity:
  Greater than 98.0% as determined by:
（a） Analysis by RP-HPLC.
（b） Anion-exchange FPLC.
（c） Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

 
Amino acid sequence:
  The sequence of the first five N-terminal amino acids was determined and was found to be
Met-Ala-Ala-Ile-Gly

 
Dimers and aggregates:
  Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

 
Biological Activity:
  ProSpec’s Human Oncostatin M is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of Human TF-1 cells is < 2 ng/ml, corresponding to a Specific Activity of 5 x 105 IU/mg.

 
Endotoxin:
  Less than 0.1 ng/µg （IEU/µg） of recombinant OSM.

 
Protein content:
  Protein quantitation was carried out by two independent methods:

1. UV spectroscopy at 280 nm using the absorbency value of 0.45 as the extinction coefficient for a 0.1% （1mg/ml） solution. This value is calculated by the PC GENE computer analysis program of protein sequences （InliGenetics）.
2. Analysis by RP-HPLC, using a standard solution of OSM as a Reference Standard.

Background :
  P-Selectin （Plaet Alpha-Granule Membrane Protein/CD62/Granulocyte Membrane Protein GRMP） belongs to a family of divalent cation-dependent carbohydrate-binding glycoproteins or adhesion molecules. Pselectin is expressed transiently on the surface of activated plaets and endothelial cells. Secreted P-selectin is thought to play a key role in the adhesion of plaets to monocytes and neutrophils during an inflammatory response. Levels of P-Selectin may be elevated in a number of pathological conditions.

 
Description :
  Recombinant Human P-Selectin is expressed in E. coli containing 566 amino acids 197-761 fused to an amino terminal hexahistidine tag.

  L-Selectin belongs to a family of divalent cation-dependent carbohydrate-binding glycoproteins or adhesion molecules. Lselectin is expressed constitutively on lymphocytes, monocytes and granulocytes and interacts specifically with carbohydrate groups on activated endothelial cells. L-Selectin may be shed by proteolytic cleavage and circulating levels in biological fluids may be used as an indicator of various pathological conditions.

经典实验操作（仅供参考）

激酶实验：

HDAC 活性实验 :
使用杆状病毒感染的Sf9细胞表达全长HDAC蛋白。此外, HDAC3共表达作为与人类NCOR2作用的复合体。为了测定含HDAC1细胞复合体活性, 免疫沉淀反应的HDAC1复合体和组蛋白H4肽段[生物素-(6-氨基)Gly-Ala-(乙酰基[3H])Lys-Arg-His-Arg-Lys-Val-NH2]的[3H]乙酰基标记的片段在50μL酶实验buffer(25mM HEPES (pH 7.4),1 M 蔗糖, 0.1 mg/mL BSA 和0.01% (v/v) Triton X-100)中温育。在37oC下温育45分钟，或者在室温下温育30分钟。加入底物之前，加入浓度不断增加的HDAC抑制剂，然后在室温下预温育10分钟。温育后，加入35μL 终止 buffer (1 M HCl 和0.4 M 乙酸)终止反应。用800μL乙酸乙酯提取释放 [3H]乙酸，然后通过闪烁计数器量化。通过Western blot 分析，测定等量的进行免疫沉淀反应的HDAC1。
