磺化Cy7是Cy7(Cyanine 7)的水溶型。它的光谱性质和Cy7类似,属近红外荧光染料。由于它的荧光波长(Em:774nm)恰好处于肌体组织近红外窗口 I 的区域(肌体的血液,体液和组织此区域背景荧光弱,而长波长穿透性强),所以磺化Cy7常常应用于小动物活体体内成像中。磺化Cy7荧光肉眼不可见,显微镜观察时需要近红外滤片,很多荧光显微镜(包括共聚焦显微镜)并不配备,但是流式细胞仪(flow cytometry)常常配备Cy7滤片组。磺化Cy7是常用荧光染料,磺化Cy7可以用来标记核酸,蛋白,抗体,多肽等,它是最常见的小动物显影剂之一。

NHS活化酯 (N-羟基琥珀酰亚胺酯,N-hydroxysuccinimide ester,succinimidyl ester)是生物标记反应中最常用的活化基团。它活化Cy7分子中的羧基,让它可以和目标生物分子上的胺基(伯胺或者仲胺)反应生成稳定的酰胺键,从而把磺化Cy7分子标记到生物大分子上。由于游离胺基是蛋白,抗体,多肽表面的常见官能团(来自于赖胺酸侧链),磺化Cy7-NHS可以和它们直接反应。核酸分子中天然不带游离胺基,只有胺基修饰过的RNA或者DNA才可以和磺化Cy7-NHS反应。

磺化Cy7-NHS活化酯的水溶性极高,所以在标记反应中不需要使用有机共溶剂,特别适合标记蛋白等对有机溶剂敏感的生物分子。另外磺化Cy7水溶性极好,并且染料分子本身带电荷,标记到蛋白表面后不会引发疏水性蛋白聚集,提高荧光标记产物的稳定性。磺化Cy7-NHS也溶于甲醇,DMSO,DMF等有机溶剂,标记反应也可以在纯有机溶剂中进行。

磺化Cy7-NHS标记后的产物Ex/Em是748/774nm。和它具有相似荧光特性的染料包括Alexa Fluor 750,DyLight 755等。

注意事项

NHS ester基团对湿度比较敏感,务必干燥保存产品。使用前产品需放到室温回温至少20分钟。

使用不含氨基的缓冲体系。比如磷酸钠缓冲液(PBS),Hepes缓冲液,碳酸氢钠缓冲液;或硼酸缓冲液等。不要使用任何含伯胺或仲胺的缓冲液,比如Tris,甘氨酸等。高浓度咪唑(imidazole)也会影响反应。

标记反应不要放在强光下,因为大部分荧光化合物在强光下长时间照射会缓慢光漂泊(photobleaching)。
